Heya! I'm trying to get started with my first tissue culture attempt. I bought some media mix: HiMedia PT080-5L Murashige and Skoog Medium with Sucrose, 6-BAP, Agar and without CaCl2, IAA, Kinetin. Perhaps this was a mistake, since you're supposed to use a 1/3 concentration of MS for VFT's.

My question is, what would you do to make VFT culture media from this mix? I plan on getting PPM and adding it as well, btw. My concern is that if I short the concentration for a 1/3 strength media, I'm shorting on the other nutrients and agar as well.

Way too strong on the hormones for a VFT even if you diluted to 1/3. VFT cultures, once started, do not need hormones for multiplication. It would be much less trouble to buy a bottle of Phytotech's Pretransplant mix C216, add sugar, pH, then add gel.
