I put the top part of a Jaws flower stalk into a culture on the 29th of May. Amazingly it started forming callus rather quickly at the place where the pedicel joins the main stalk and in other places. I think I noticed the little nubs forming about 7 or 8 days after putting it into the culture.

Unfortunately, the flower buds were contaminated, so I had to pull the tissue out of the culture a couple days ago and snip off the buds and resterilize. Seems to be doing fine and the callus is progressing!

Nice work! Are you still using the same media as before? Any different conditions?

Nope, same process just a different plant part harvested at a different time. It seems like (at least for me) that the top of the flower stalks is the fastest to form callus (a week!), follow by leaves (4 weeks), followed by the main part of flower stalks (almost never!).

My media is the standard 1/2 MS with full strength vitamins, 30g of sugar, and 1m PPM with a pH of 5.6. I've decided not to use BAP or any other cytokinin for callus induction as VFTs don't seem to need it and it can cause the tissue to turn brown much quicker.

Wow Matt! Looks awesome, interesting info on the time it takes for the different parts of the plant to callus. question about leaves tho, how small of a cutting on leaves can you get away with without killing all the tissue?

Hi Matt,

I have the same kind of tissue in culture from my Royal Red flower bud, planted on the same date. I too have noticed some red colour formations, besides the fact that mine is a Royal ‘Red’. Unfortunately as time passes some sort of ‘grey/white film’ is showing on my medium. There is no sign of mold, but maybe it’s bacteria growing. It has not fully covered my tissue (it only started two days ago). I probably should replate it again, but how do I sterilize? I used a harsh sterilisation technique before bringing my tissue in culture. Mechanical removal under running water, washing with water containing tween 20 for 20 minutes, 20 min 10% bleach, washes with sterile aqua dest en 5 min wash with 10% H2O2 (30% H2O2 diluted to 10%). Should I do the same sterilisation again with this callus forming tissue? My medium consists of activated charcoal as described in an article you mentioned in your blog, "Source, etiolation and orientation of explants affect in vitro regeneration of Venus fly-trap (Dionaea muscipula)" by W.L. Teng in 1998. What are your suggestions about sterilizing and media for replating this piece?

Hi swerfer,

Just a suggestion in replating. If you don't see any signs of contamination and it's usually the case, you can just replate the tissue and divide it if you will in plain 1/3 MS, 30 grams of sugar/liter.
