Hey guys,

got some problem which appears from time to time. A plant in tissue culture begins to get brownisch and dies off if it not gets replated very soon. If i dont replate this plant dies within one week. If i open the vessel then there is a vacuum an i hear the air going in when pulling up the lid. Then the medium changes the color from clear to yellowish. This appears within hours and at the next day the whole media is yellowiah after contact with fresh O2.

I linked a photo with shows excactley the coloration of plants. For me it is mostly VFF.
Do you have an idea what causes this phenomenon?

It kind of sounds like bacterial contamination except the plant should continue dying after re-plating if that's the case. How is it that you're achieving a vacuum/low pressure in the tube?

Sometimes bacteria can cause that. But it will often happen with clean cultures as well. Seems to be due to the plant secreting something that poisons itself.

Some varieties of flytrap excrete a lot of it because I have had trouble growing quite a number of varieties in TC over the years. They brown so quickly after a replate that it requires constant new media or they die.

Other times it is due to damage caused to the tissue during replate. If the plant tissue is damaged, it seems to excrete these chemicals at a very high rate.
