I've got a Cephalotus follicularis today.


And I want to start tissue culture of it from cutting leaves.
I've already setup the medium (1/5 MS suger:20g/L agr:6g/L +little BAP and little NAA)

I have some questions.

May I cut a leaf into small pieces ?
or
Dose it need whole leaf to grow ?

I want to try by flat leaves.

I'm afraid : I am not good at English, So I will ask something stupid. Please understand it.

The problem I had when I tried was effectively sterilizing them without killing them. So a larger leaf might hold up to your sterilization better than smaller pieces. If I tried again I would probably try sterilizing in PPM or PTC3. But sorry, I don't know which will yield better growth since I never succeeded to get one started.

I tried to sterilize by 75% ethanol for 30 seconds then soaking in 50ppm Hypochlorous acid solution.
and rinced it well in water.

If this method is too strong for leaf, I will try PPM or other .

I'm in Japan ( ^ . ^ )/ I'm Japanese.
