Manually? No. I rinse out pots and let them sit in the sun for at least a week before reuse so the sun's UV rays can take care of that.

Yes I do mean the baby food containers or whatever you are using for TC. Thanks Panman for pointing that out.

Yes, you want to sterilize the TC container before use. Otherwise, bacteria and/or mold will wreak havoc on your plants and media.

Always sterilise just about everything going into tissue culture. Here at uni I use the autoclave @121c for 20 minutes. At home you can use a pressure cooker to do so, just don't seal your containers entirely or they may explode!
