Hi ,
I am looking for a methods to sterilize Nepenthes seeds for in vitro work.
I have tested bleach but the results is not so good.
Has anyone that knows a better methods and may teach me.

Hello Marcos!

what type of train of desinfection are you usign?
at what % the chlorine are you using?

they use 15% Clorox (bleaching solution) and
0.01% tween 80 (v/v) for 20 min. and again with 5% Clorox for 10 min, then washed three times with sterilized distilled water for 3 min. each.

I havent put my hands on nephentes seeds but if I do I will do this.

also it is better if the seeds are still on the pod.

Than you very much for your help. Sorry for so late unswer.
This method seems to kill all seeds.
I tryed 10% an 5% for 10 min, 5 min but no good answer .

Hi guys

Read my method on Hello today Here is a copy. Use 70% Ethanol for 30 sec then wash in SDICN one tablet Milton stock solution (800 mg in 100 ml water) for 10 minutes. No need to wash the seed before plating just squirt the seed and SDICN water onto 1/3 MS . The MS media you can make on the Kitchen bench. Add all ingredients including Agar etc heat to 91oC stirring then allow to cool to 51oC add 20 ml of the Milton stock solution then dispense into the cheapest flasks you can find. There is no need to autoclave any thing just clip down the lid once the media sets Good for 6 weeks The chlorine will sterilise everything inside. Believe me it works. Have a try
