caramelization media occurs when u autoclave too long subjecting high heat to sugar components of the media.Sugars tend to degrade and form melanoidins which are brown and have high molecular weight compounds.They are responsible for growth inhibition during culturing of tissues.This can be prevented by filter sterilization of carbon source say sucrose and thoroughly mixed to media after autoclave.
