PLB and cellus

Medium for make:

*PLB (protocorm-like body): MS1/2 (1/3) + sucrose 30g/l, NAA 1mg/l, BA 10mg/l.

*Cellus from PLB: MS 1/2 (1/3) + sucrose 30g/l, NAA 0.3mg/l, BA 1mg/l.

*explant medium: MS 1/2 (1/3) + sucrose 30g/l, NAA 0.5mg/l, BA 0.5mg/l,

That is the general medium, now I want to replace BA to BAP (I just have BAP now :lol: ), is it OK?

Anyone try this medium yet?

BA or BAP is the same: 6-BENZYLAMINOPURINE. The formula are for CPs?

That is good to know, my friend told me they are different, haha!

That formula for orchid (only use MS or MS 1/2). I just change MS concentration.

Now I planning to try make PLB from nepenthes seeds (or leave) and VFT seeds.

You're better to ask Geoff or Matt about hormones for CPs. Each plant prefers different amount and hormone. However, it's not hurt to try different formula if you have extra explants. It's fun to try. I always do the same. :)

PLB is more associated with orchid terminology, Aaron can explain this way better than me.
I have not come across any cp that can tolerate 10mg BAP, 5-7mg seems to be the max and I would never place seed on this.
By all means experiment, this is what it is all about. Tc is an evolving science it is always moving forward, or backwards :? . Hopefully we learn from our mistakes.

Hi Geoff,

That mean PLB medium only use for explant (nep and FVT)?

May I ask a stubid question :D 7mg/l BAP = 7ml BAP (1g/l)?

Yeah- I think the more appropriate term would be to form callus, rather than PLBs, when speaking about anything other than orchids. And when the callus just falls apart at the touch of a tool- no cutting required- it's called friable callus.

Hi ahicks51, did you try it before? Now I need some advise from predecessors!

I am trying MS 1/3 + sucrose 30g/l + 1mg BAP, 0.3mg NAA

Is it medium work ok? Can it make PLB or cellus?
