What I have to add to my next media to generate roots?

Nothing, just use spaghum moss and place it ontop. you should also keap it lightly wet, so it doesnt rot. also if you have a place with a high humidity and indirect sunlight, place it there. If you dont, make the humidity high by covering it with a plastic bag. Have a few holes in the bag for ventilation, and after you want to place it back into normal conditions slowly make more and more holes into the bag over a few days until the humidity is back to normal.

Perhaps the medium, or something you're adding to it has hormones to prevent root growth, not sure why you'd want to get roots while it's in culture, but i'm not an expert

I want to transfer to another media but I need to induce roots, how can I prepare this new media?

To induce root:
- Add 1mg/l NAA into medium.
- Add 1 - 2 mg/l charcoal. Some species need dark medium to grow root.
- Root growth is often depressed by NH4+ and promoted by NO3. Reduce N increase P. Choose another medium or adjust your medium .
- Add 0.05% - 0.1% Casein hydrolysate.
Some seedlings are hard to get root in vitro but get good root when deflask.
I usually add 1mg/l NAA for convenient, a little bit better.

I don't have Casein hydrolysate...do you think that I can try only with NAA and Charcoal?

Sure, use NAA or charcoal or both at the same time. Try and watch is the name of the game.

Ok, I have to make 500ml of media to induce roots. Please tell me if I'm wrong with this process:


1.1g MS with macro+micro+vitamins
15g of suggar
0.5ml of PPM
0.5mg NAA
1mg charcoal
Agar

Should I use BAP?Kinetin? IAA?
